propagation medium: 90% RPMI 1640 + 10% FBS + 2 mM L-glutamine + 30 U/ml rh or rm IL-2 (CTLL units) maintain at 5-30 x 104 cells/ml; split ratio 1:3 to 1:4 every 3 days; cells have to be feedered every two weeks with spleen cells of H-2 k mice; feeder medium: 90% RPMI 1640 + 10% FBS + 2 mM L-glutamine + 10% (vol) supernatant of Con A-stimulated mouse spleen cells, 0.05 M methyl-alpha-D-mannopyranoside, 200 ęg/ml conalbumin; seed out at ca. 3 x 105 cells/ml at 37C with 7.5% CO2 maximal density at 0.6-1.0 x 106 cells/ml;
doubling time of ca. 30 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1.5-2.0 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: immunological analysis (half maximal proliferation with about 10pg rh IL-1a/ml in MTT assay)
