Description: Swiss albino mouse fibroblasts established from disaggregated Swiss albino mouse embryos in 1962; cells are used for transfection studies with DNA viruses and as assay system for transformation studies; cells have a high sensitivity to contact inhibition (= ATCC CCL 92) confirmed as mouse with IEF of AST, LDH, PEP B

90-95% Dulbecco's MEM + 5-10% FBS split confluent culture at a ratio of up to 1:8 every 2 to 4 days using trypsin/EDTA; after trypsination cells might remain in aggregates that are difficult to disperse; upon reaching confluence cells stop to grow; cells grow fast at low titer; seed out at ca. 1-2 x 106 cells/175 cm 2 at 37C with 5% CO2 cell harvest of about 5-8 x 106 cells/175 cm 2;
doubling time of ca. 40 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at 1 x 106 cells/ampoule;
mycoplasma negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (murine hypertriploid (near tetraploid sideline present) karyotype with 13% polyploidy; 68(58-73)<3n>) * isoenzymes (confirmed as mouse with IEF of AST, LDH, PEP B)
