80% RPMI 1640 + 20% FBS split 1:2 to 1:4 every 6-10 days using trypsin/EDTA; initial growth after recovery is slow; seed out at ca. 1-2 x 106 cells/80 cm 2 at 37C with 5% CO2 cell harvest of about 1.5 x 105 cells/cm 2;
doubling time of about 50-100 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (apparently human karyotype) * DNA fingerprinting (unique DNA profile with (gtg)5 multilocus probe) * immunological analysis (cytokeratin+, desmin-, endothel-, GFAP-, neurofilament-, vimentin-) * isoenzymes (confirmed as human with IEF of AST, MDH) * reverse transcriptase (reverse transcriptase not detected)
