90% RPMI 1640 + 10% FBS maintain at 0.5-2.0 x 106 cells/ml, split 1:2 to 1:3; seed out at 0.5 x 106 cells/ml at 37C with 5% CO2 saturation density of over 2 x 106 cells/ml;
doubling time of about 35 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (near diploid with about 3% triploidy; a supernumerary dmin was present in about 25% metaphases; 46(45-48)<2n>X/XY, dup(1)(q21q32), dup(7)(q12q22), t(8;14)(q24;q32); additional rearrangements were present in a sideline, viz.t(6;1 )(p2) * cytogenetics 2 (and del(11)(p11)) * DNA fingerprinting (unique Hinf I-(gtg)5 DNA profile) * immunological analysis (CD3- CD10+ CD13- CD19+ CD37+ HLA-DR+ sIgM+) * isoenzymes (confirmed as human with IEF of AST, MDH, NP)
