Coon's modified Ham's F12 + 2mM Glutamine + 5% Fetal Bovine Serum (FBS). Split semi-confluent cultures (70%) 1:10 i.e. seeding at 9x1000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C. Saturation density is reached at 15x1,000,000 cells/10cm² dish.

C2-Rev 7 is a revertant of the C2 variants, selected by virtue of its capacity to grow in glucose free medium (containing dialysed serum) where survival requires activity of the liver-specific gluconeogenic pathway.
