92.5% Dulbecco's MEM (4.5 g/L glucose) + 5% horse serum + 2.5% FBS maintain at 0.5-1.0 x 106 cells/ml; optimal split ratio of 1:2 every 2-3 days at 37C with 5% CO2 maximum density at about 1-2 x 106 cells/ml;
frozen with 70% medium, 10% horse serum, 5% FBS, 10% DMSO at about 5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (92(88-101)<4n>; murine hypertetraploid karyotype with centric fusion and 1.7% polyploidy) * isoenzymes (confirmed as mouse with IEF of AST, NP) * reverse transcriptase (reverse transcriptase negative)
