80% RPMI 1640 + 20% FBS + 10 ęg/ml bovine insulin + 2 mM L-glutamine split confluent culture 1:2 to 1:3 every 7-10 days using trypsin/EDTA; cells grow slowly; seed out at ca. 2-3 x 106 cells/25 cm 2 at 37C with 5% CO2 about 1.0-1.6 x 105 cells/cm 2 or 20 x 106 cells/175 cm 2;
doubling time of ca. 100 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1-2 x 106 cells/ampoule;
contamination was eliminated with Mycoplasma Removal Agent, then negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
