45% Ham's F10 + 45% Dulbecco's MEM + 10% FBS + 5 æg/ml bovine insulin + 5 æg/ml human transferrin + 50 nM hydrocortisone + 5 ng/ml selenite + 10 æM HEPES split confluent culture 1:4 every 6-7 days using trypsin/EDTA at 37C with 5% CO2 harvest of 2-3 x 106 cells/80 cm 2;
doubling time of about 60-75 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1-2 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (murine hypotetraploid karyotype with 14% polyploidy and 1-3 d-min in 34% cells; 70(64-74)<4n>; 1-3 centric fusion markers present) * isoenzymes (confirmed as mouse with IEF of AST, NP) * reverse transcriptase (reverse transcriptase not detected)
