80-85% RPMI 1640 + 10-15% FBS + HFCS ('Hybridoma Fusion and Cloning Supplement') for the first 1-2 weeks post-thawing cells are difficult to culture! maintain at ca. 0.2-0.8 x 106 cells/ml; split saturated culture 1:2 to 1:4 every 2-3 days; seed out at ca. 0.2 x 106 cells/ml; after thawing cultivate cells for the first 1-2 weeks in 24- or 12-well-plates using HFCS at 37C with 5% CO2 maximal density at ca. 0.8 x 106 cells/ml;
doubling time of ca. 35 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 4-5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (murine karyotype) * immunological analysis (secretion of a mouse IgG2 monoclonal antibody; we suggest to subclone the culture as about 3% of the cells reacted strongly with FITC anti-mouse IgM) * isoenzymes (confirmed as mouse with IEF of AST, MDH, PEP B) * reverse transcriptase (reverse transcriptase not detected)
