This is the complete description of the cell line that you requested.
By selecting a generic term (e.g. SPECIES) you will go to the corresponding list of controlled terms, while by selecting a specific term you will go to the list of corresponding cell line names.
SPECIES: human
TUMOR: neuroblastoma
DSMZ catalog code: ACC 157
Morphology: adherent, fibroblast-like cells growing as monolayers
Description: human neuroblastoma established from a neuroblastoma metastasis at an adrenal site of a 4-year-old white boy in 1986; cells were described to express neuron-specific enolase and synaptophysin (but neither GFAP nor S-100 protein) and to carry a 20-fold amplification of N-myc confirmed as human with IEF of AST, MDH
Viruses: ELISA: reverse transcriptase negative; PCR: EBV-, HBV-, HCV-, HHV-8-, HIV-, HTLV-I/II-
Depositor: Dr. T. Pietsch, University of Bonn, Institute of Neuropathology, Bonn, Germany
General Restrictions
Available in the following LABORATORY:
-
German Collection of Microorganisms and Cell Cultures (DSMZ, Braunschweig)
90% RPMI 1640 + 10% FBS + 2 mM L-glutamine + MEM non-essential amino acids split confluent culture 1:2 every 4 days using trypsin/EDTA; NB: medium must be changed frequently to prevent acidity which induces cell clumping; optimal dissociation after trypsination is affected by gentle aspiration with a 21 gauge needle and syringe at 37C with 5% CO2 cell harvest of about 10 x 106 cells/80 cm 2;
doubling time of ca. 100 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1-2 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (human bimodal hyper, -diploid/-tetraploid karyotype; 54(48-54)<2n>XY, +1, +1, +2, +6, +7, +8, +13, +20, der(1)t(1;?)(p32;?)x2, der(13)t(13;2HSR)(q33;?p23); both the long HSR on ch13 due to N-myc amplification and the ch1 short-ar) * cytogenetics 2 (rearrangements are typical of neuroblastoma) * immunological analysis (cytokeratin -, desmin -, endothel -, GFAP -, neurofilament +, vimentin +) * isoenzymes (confirmed as human with IEF of AST, MDH) * reverse transcriptase (reverse transcriptase negative)
