Coon's modified Ham's F12 + 2mM Glutamine + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:3 to 1:4 i.e. seeding at 3-4x10,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37C.

Derived from normal rat lung tissue. The cells possess elevated levels of specific enzymes involved in the synthesis of phosphatidyl choline. Osmiophilic lamellar inclusions are observed in cell pellets from mass culture.
