Description: mouse monocytes-macrophages established from a tumor in a female BALB/c mouse in 1968; cells were described to synthesize lysozyme and interleukin-1 and to have receptors for immunoglobulin and complement confirmed as mouse with IEF of AST, MDH, NP, PEP B

90% Dulbecco's MEM + 10% FBS split confluent culture 1:3 to 1:4 about twice a week (prior to reaching confluence); cells detach spontaneously if allowed to exceed confluence; cells may be detached with a rubber policeman or using TNE (40 mM TrisCl pH 7.5, 1 mM EDTA pH 7.5, 150 mM NaCl) (not trypsin/EDTA) for quantitative harvesting or for seeding out of cells collected from the supernatant for passaging; use always new culture flasks, otherwise cells are difficult to detach; seed out at ca. 1-2 x 106 cells/80 cm 2 at 37C with 5% CO2 about 4.5 x 107 cells/80 cm 2;
doubling time of ca. 35 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (murine hypotetraploid karyotype; 78(70-81)<4n>; in most cells a centric fragment was present) * isoenzymes (confirmed as mouse with IEF of AST, MDH, NP, PEP B)
