80% Dulbecco's MEM + 20% FBS cell growth is very slow! split confluent culture 1:2 to 1:3 once a week using trypsin/EDTA; seed out at ca. 0.5-0.8 x 104 cells/cm 2 at 37C with 5-10% CO2 about 1-2 x 106 cells/80 cm 2;
doubling time of ca. 50-100 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1 x 105 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: DNA fingerprinting (unique Hinf I-(gtg)5 DNA profile) * isoenzymes (confirmed as rat with IEF of AST, MDH, NP) * reverse transcriptase (negative)
