90% RPMI 1640 + 5% FBS + 5% ConA spleen cell supernatant (or 25 U/ml IL-6; or 10% vol conditioned medium of cell line 5637 (DSM ACC 35) + 2.5 x 10-5 M mercaptoethanol + 2 mM L-glutamine maintain at 0.1-0.3 x 106 cells/ml, optimal split ratio (daily) of 1:3, maximal split ratio of 1:5 at 37C with 7.5% CO2 maximum density of 0.5-1.0 x 106 cells/ml;
doubling time of ca. 24-25 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 5 x 105 cells/ampoule;
contamination was eliminated with Ciprobay (ciprofloxacin), then negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: immunological analysis (convenient bioassay for IL-6) * isoenzymes (confirmed as mouse with IEF of MDH, NP, PEP B)
