80% RPMI 1640 + 20% FBS + 2 mM L-glutamine + 1x MEM non-essential amino acids + 1 mM sodium pyruvate split confluent culture once a week using trypsin/EDTA; seed out about 1-2 x 106 cells/175 cm 2 , after thawing cells might at first grow very slowly at 37C with 5% CO2 cell harvest of about 6 x 106 cells/175 cm 2;
doubling time of about 43 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 0.5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (apparently human karyotype) * DNA fingerprinting (unique DNA profile with (gtg)5 multilocus probe) * immunological analysis (cytokeratin+ (100%) desmin- endothel- GFAP- neurofilament- vimentin+ (100%)) * isoenzymes (confirmed as human with IEF of AST, MDH) * reverse transcriptase (reverse transcriptase negative)
