90-95% RPMI 1640 + 5-10% FBS split confluent culture 1:4 to 1:5 two to three times per week using trypsin/EDTA; seed out at ca. 1-2 x 106 cells/80 cm 2 at 37C with 5% CO2 cell harvest of about 5 x 106 cells/80 cm 2;
doubling time of ca. 48 hours (with 5% FBS);
frozen with 70% medium, 20% FBS, 10% DMSO at about 1 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
