EMEM (EBSS) or TC-199 + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Fetal Bovine Serum (FBS). Spilt confluent cultures 1:3 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 25-28C.

The cell line CLC (Carp Leukocyte Culture) was established from peripheral blood mononuclear cells of a normal 2-year-old female common carp, Cyprinus carpio. They possess phagocytic activity and are reported to be stimulated by TNF, gamma-interferon and LPS therefore resembling characteristic features of monocytes / macrophages. CLC replicate fish viruses e.g. IPNV, PFR, SVCV and VHSV.
