Part of the Business Division of the Health Protection Agency (ECACC, Salisbury, Wiltshire)
EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:3 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 30C.
CCO was established from the ovaries of a healthy, juvenile channel catfish, Ictalurus punctatus. The cells grow at temperatures from 10C to 35C with the optimum at 30C and with a reported population doubling up to 12 hrs. CCO cells replicated channel catfish virus but were refractory to 12 other viruses from fish, birds and mammals.
