Description: mouse hybridoma (anti-fiber/myosin) described as secreting a mouse monoclonal antibody (IgM) reacting with slow muscle fibers in rodent, chicken and human and with slow myosin heavy chain on Western blots; the cell line was established in 1985 by fusion of X63AG8.653 cells with spleen cells from a BALB/c mouse that had been immunized with human skeletal muscle myosin confirmed as mouse with IEF of AST, NP

90% Dulbecco's MEM (4.5 g/L glucose) + 10% FBS + 2 mM L-glutamine + 1 mM sodium pyruvate + 1x non-essential amino acids split saturated culture 1:5 every 3 days; seed out at about 0.3 x 106 cells/ml at 37C with 5% CO2 maximal density of about 1.5 x 106 cells/ml;
doubling time of about 25 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 2 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: immunological analysis (cytokeratin+ desmin- endothel- GFAP- neurofilament- vimentin-) * reverse transcriptase (reverse transcriptase negative)
