Morphology: single, round cells in suspension (some cells adhere partly, but trypsin is not necessary to detach these cells)
Description: mouse hybridoma (anti-rat fiber/myosin) this hybridoma cell line was described as secreting a mouse monoclonal antibody (IgG 1 ) that reacts with rat neonatal fibers and, to a lower extent, with all rat adult type 2 fibers in immunofluorescence; it reacts specifically with neonatal MHCs (myosin heavy chain) in Western blotting; established in 1984 by fusion of NSO myeloma cells with spleen cells from a BALB/c mouse immunized with purified myosin from fetal bovine muscles confirmed as mouse with IEF of AST, MDH, PEP B

90% Dulbecco's MEM (4.5 g/L glucose) + 10% horse serum + 2 mM L-glutamine split saturated culture at 1:2 to 1:3 every 2-3 days at 37C with 5% CO2 maximal density at about 1.5-2.0 x 106 cells/ml;
doubling time of ca. 30-40 hours;
frozen with 70% medium, 20% horse serum, 10% DMSO at about 5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (murine chromosome morphology) * immunological analysis (secretion of a mouse IgG1 monoclonal antibody) * isoenzymes (confirmed as mouse with IEF of AST, MDH, PEP B) * reverse transcriptase (reverse transcriptase not detected)
