90% Medium 199 (25 mM HEPES) with Earle's salts with L-glutamine + 10% FBS cell growth is extremely slow; split confluent culture 1:2 every 7 days using TNE (50 mM Tris, 0.1 M NaCl, 5 mM EDTA, pH 7.5) at +4C for 10 min; culture at room temperature (about 20C) without CO2 (keep lid of flask closed); cells can be stored temporarily (up to 1 week) at 4C; seed out at ca. 0.1 x 106 cells/ml at room temperature (about 20C) without CO2 about 5 x 106 cells/175 cm 2 - saturation density of ca. 10 x 106 cells/175 cm 2;
doubling time of about 160 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 3 x 106 cells/ampoule;
contamination was eliminated by the depositor with BM-Cyclin (tiamulin & minocycline), then negative in DAPI, microbiological culture, RNA hybridization assays;
