This is the complete description of the cell line that you requested.
By selecting a generic term (e.g. SPECIES) you will go to the corresponding list of controlled terms, while by selecting a specific term you will go to the list of corresponding cell line names.
SPECIES: human
TUMOR: leukemia, acute monocytic
DSMZ catalog code: ACC 102
Morphology: single, round cells growing in suspension
Description: human acute monocytic leukemia established from a 10-year-old boy with acute monocytic leukemia (AML FAB M5) at diagnosis; cells were described to carry a 4;11 translocation; patented cell line confirmed as human with IEF of MDH, NP, PEP B
Viruses: ELISA: reverse transcriptase negative; PCR: EBV-, HBV-, HCV-, HHV-8-, HIV-, HTLV-I/II-
Depositor: Dr. H. G. Drexler, DSMZ, Braunschweig, Germany
General Restrictions
Available in the following LABORATORY:
-
German Collection of Microorganisms and Cell Cultures (DSMZ, Braunschweig)
90% RPMI 1640 + 10% FBS maintain at 0.2-1.0 x 106 cells/ml; split carefully; do not exceed a cell density of about 1 x 106 cells/ml; cells are difficult to culture and grow slowly at first; addition of recombinant GM-CSF or IL-3 (or as 10-20% vol conditioned medium of cell line 5637, DSM ACC 35) or increasing the FBS content to 20% can be of advantage; cultivation in 24-well-plates appears to be better than in large culture flasks at 37C with 5% CO2 saturation density at about 1.0 x 106 cells/ml;
doubling time of ca. 50 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (hyperdiploid; 48(46-48)<2n>XY, +8, +18, +19, -21, t(4;11)(q21;q23)) * DNA fingerprinting (unique Hinf I-(gtg)5 DNA profile) * immunological analysis (CD3- CD4+ CD5- CD8- CD10- CD13+ CD14- CD15+ CD19- CD21- CD25- CD33+ CD37- DR+) * isoenzymes (confirmed as human with IEF of MDH, NP, PEP B) * reverse transcriptase (negative for HTLV-I/II, HIV-1)
