General Restrictions
Available in the following LABORATORY:
-
German Collection of Microorganisms and Cell Cultures (DSMZ, Braunschweig)
80-90% Iscove's MDM + 10-20% FBS maintain at 0.5-1.0 x 106 cells/ml, cells grow slowly, split 1:2 every 2-3 days; viability may be low initially after thawing; it is recommended to cultivate the cells in 24-well-plates during the first week and to exchange medium without dilution; seed out at ca. 0.7 x 106 cells/well at 37C with 5% CO2 maximal density at about 1.0-1.5 x 106 cells/ml;
doubling time of 35-80 hours (depending on cell density);
frozen with 70% medium, 20% FBS, 10% DMSO at about 5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (hypertriploid karyotype; 79(75-79)<3n>XX, -X, complex karyotype including LP-1 markers: dup(1)(p13;p23), der(5)t(5;?)(q31;?), der(12)t(12;13)(p13;q11); in 33% cells 1, 2 or occasionally 4 supernumerary dm present) * DNA fingerprinting (unique Hinf I-(gtg)5 DNA profile) * immunological analysis (CD3- CD13- CD19- CD37- HLA-DR+ sIgG+) * isoenzymes (confirmed as human with IEF of AST, MDH, NP) * reverse transcriptase (reverse transcriptase negative)
