Part of the Business Division of the Health Protection Agency (ECACC, Salisbury, Wiltshire)
EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37C.
Karyotype: Hyperdiploid
This cell line was selected by passaging A9 (ECACC No. 85011426) through syngenic C3H or semi allogenic mice over a period of three months and uptake was considerable higher than A9, thus A9 high uptake or A9HT. Karyotype is similar to A9 but with a slightly reduced total number of chromosomes, with the modal number of 53. This line lacks inosinic acid pyrophosphorylase.
