85-90% McCoy's 5A + 10-15% FBS split confluent culture 1 x per week at up to 1:5 using trypsin/EDTA; seed out directly in a 80 cm 2 culture flask, cells will reach confluency within 5-8 days; cells may clump after trypsinization and may be difficult to disperse at 37C with 5% CO2 cell harvest of about 5-10 x 106 cells/80 cm 2; doubling time of about 36 hours; frozen with 70% medium, 20% FBS, 10% DMSO at about 1-2 x 106 cells/ampoule; negative in DAPI, microbiological culture, RNA hybridization assays;
