Morphology: epithelial cells growing as monolayers
Description: human cervix carcinoma (derivative of HELA) these cells were cloned from the GIRARDI HEART cell line (ATCC CCL 27, passage 623) in 1981 and were thought to 'represent human heart cells'; however, isoenzyme analysis (Nelson-Rees et al., Science 191: 96-97, 1976) and DNA fingerprinting at the DSMZ established unequivocally that this cell line must be considered de facto a subclone of HELA; cells were described to be sensitive to various toxins and hormones confirmed as human with IEF of AST, MDH

45% Medium 199 (with Hank's salts) and 45% RPMI 1640 + 10% FBS split confluent culture 1:3 to 1:6 every 3-4 days using trypsin/EDTA at 37C with 5% CO2 about 10 x 106 cells/80 cm 2;
frozen with 70% medium, 20% FBS, 10% DMSO at about 2-3 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: DNA fingerprinting (same Hinf I-(gtg)5 DNA profile as HELA) * immunological analysis (cytokeratin+ desmin- endothel- GFAP- neurofilament- vimentin+) * isoenzymes (confirmed as human with IEF of AST, MDH) * reverse transcriptase (negative in reverse transcriptase assay)
